Enhancement of CD8 T cell response upon exposure to autologous DC loaded with low numbers of apoptotic CTCL cells. A – D. DC loaded with 1.0 × 106 and 0.25 × 106 apoptotic malignant T cells stimulated increased perforin activity in the CD8 T cell compartment, as compared to DC co-cultured with 1.0 × 106 and 0.25 × 106 viable CTCL cells. Results are presented as quadstats measuring the expression of membrane CD8/cytoplasmic perforin as determined by flow cytometric analysis of the gated lymphocyte population. Results are representative of 5 separate experiments performed on cells isolated from cultures of 5 different CTCL patients. E. Increased perforin activity in the CD8 T cell compartment translated into increased CTCL cell death as measured by membrane CD3/cytoplasmic APO 2.7 staining. Data analyzed using student's t test. Apoptotic = CD3-treated, Viable = CD4-treated, NS = Not significant.
